Description Potently inhibits tetrodotoxin (TTX)-sensitive voltage-gated sodium (Nav) channel currents in adult rat dorsal root ganglion neurons. No effect on TTX-resistant Nav channels or on voltage-gated calcium channels. No effect observed after injection of large doses (50,156 pmol/g = 200 µg/g) in American cockroaches (Periplaneta americana). Potently inhibits human Nav1.7 and rat Nav1.2. Moderately potent inhibitor of rat Nav1.3 but no effect on rat Nav1.4 or human Nav1.5. Binds to extracellular S3-S4 linker in the domain II voltage sensor of hNav1.7 and stabilises closed state of channel.
